Abstract


The used the 277 bp partial fragment was used to isolation the full-length of the sesame polygalacturonase gene. The amplification of the 5’ and 3’ ends was carried out using 3’, 5’ RACE kit, following the manufacturer protocol. The 3’ end of PG gene was obtained by two RACE reactions outer and inner using the PG- 3’Outer/RACE outer and PG-3’Inner/RACE inner primers. This step resulted in amplification of a 639 bp fragment (PS2), and the amplification of 632 bp fragment (PS3). From the 5’ reaction, a 314 bp fragment (PS4) was obtained using PG-5’Outer /RACE outer and PG-5’Inner/ RACE inner primers.
