Evaluation of Addition of Silver Nano Particles to Common Root Canal Irrigants on Root Canal Dentine, Antibacterial Efficacy and Cytotoxicity

Abstract


This study was designed to evaluate the effect of addition on nano-silver particles to common endodontic irrigants on some root canal dentine properties as well as its antimicrobial efficacy and cytotoxicity. Freshly extracted central incisors were used. The tested irrigants were nano-silver (0.01%), NaOCl (5.25%), CHX (2%), EDTA (17%) and/or after addition of nano-silver particles to each irrigant.
The dentine properties studied were push-out bond strength, dentine permeability using confocal laser scanning microscopy, effect of irrigant on smear layer removal using scanning electron microscope and image analysis, and dentine wettability by measuring the contact angle of the irrigant. The antibacterial effect of the irrigant was measured against Enterococcus Faecalis. This was done by measuring the minimal inhibitory concentration of each irrigant to E. Faecalis and by measuring the count of Live/Dead bacteria. In which the samples were stained by Acreditine Orange dye. Specimens were imaged using confocal laser scanning microscope and image analysis was done using Zeiss Black software to count the number of dead and living bacterial cells. Cytotoxicity of the irrigants was assessed using MTT assay by monitoring the effect on baby hamster kidney fibroblasts.
