Potential Role of Annona squamousa Linn Extract or/and N-Acetyle Cysteine against Anti-Tuberculosis Drugs Causing Hepatotoxicity in Male Rats

Abstract


Drug-induced liver toxicity may be a common reason behind liver injury. It accounts for about one-half of the cases of acute liver failure and mimics all types of acute and chronic diseases. Rifampicin (Rif) is a recognized therapeutic and preventive agent against tuberculosis. However, high rates of many side effects and symptoms associated with hepatotoxicity have identified throughout the treatment. The current study was aimed to evaluate the antioxidant and hepatoprotective activity of methanolic extract of Annona squamosa Linn (MEAS) (500 mg/kg b.wt.), or/and N-Acetyl Cysteine (NAC) (100 mg/kg b.wt.) against Rif-induced hepatic injury (300mg/kg b.wt.) in male rats, orally and daily for 21 days. The results showed that Rif increased significantly the levels of bilirubin, alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), γ-Glutamyl transferase(GGT), malondialdehyde (MDA), interleukin-6 (IL-6) and 8-hydroxy-deoxyguanosine (8-OHdG) and decreased significantly the concentrations of total protein, albumin, hepatic reduced glutathione (GSH), the levels of interleukin-10 (IL-10) and B-cell CLL/lymphoma 2 (Bcl2) in Rif-treated rats.
