BIOLOGICAL AND MOLECULAR STUDIES FOR THE DETECTION OF PEA SEED BORNE MOSAIC VIRUS

Abstract
Pea Seed borne Mosaic Virus (PSbMV) is considered one of the important seed borne VIruses in Egypt. The present study was carried out to isolate and characterize the virus. The virus was isolated from commercially pea seed lot cu. Little Marvel and the resulted seedlings which showing the mosaic, leaf down folding were tested by indirectc ELISA for PSbMV presence. PSbMV bearing-seedlings were further . used for mechanical inoculation to healthy seedlings. The virus was obtained in a pure form using single local lesion technique developed on Ch. Amaranticolor. The virus was identified as PSbMV according to the symptoms developed on indicator plants, serological tests (ELISA), physical properties, mode of transmission, Electron Microscopy, Polymesrase Chain Reaction (PCR), nucleic acid hybridization and nucleotide sequence.
The obtained results were summarized as follows:
1- The host range of PSbl\-1V was found to be restricted in Fabaceae and Chenopodiaceae; symptoms were as follows necrotic local lesions on Ch. Amaranticolor, Ch. Album, and Ch. quinoa; Systemic chlorosis and leaf rolling on Vicia faba and mild mosaic, leave down folding and mild stunting of stems on Pisum Sativum cv. Little Marvel.
The obtained results were summarized as follows:
