Pathological studies on the protective Effect of Purslane(Portulaca Oleracea) Seeds Extract on Acrylamide Induced Toxicity in Rats

Abstract


This study aimed to investigate the toxic effect of acrylamide in nervous and reproductive system of rats through molecular, biochemical, pathological and immunohistochemical examinations. Also to evaluate the protective effects of portulaca oleracea seed hydroethanolic extract on acrylamide -induced toxicity.
The P. oleracea seeds were purchased from the local market (Haraz, Cairo, Egypt). Briefly, 250 grams of powdered seeds were blended with 70% ethanol in a sealed glass vessel. Then the mixture was allowed to settle down for 72 hours, in the dark at room temperature, and then was filtered. After completion of maceration (3 times), the solvent was evaporated under vacuum using a rotary evaporator (Heidolph, 60 rpm Laborta 4000) at a temperature below 50°C and RPM 60 to yield a viscous extract. The obtained extract was stored at -20°C until used.
PSE was subjected to phytochemical investigation using ultra-high-performance liquid chromatography (UPLC) coupled with quantitative time of flight mass spectrometry (qTOF-MS).
In this study UPLC-MS fingerprinting of the PSE extract showed Sixteen Octadecanoids (C18) fatty acyls with different oxidation and unsaturation level could have been tentatively identified including phytofuran and phytoprostane derivatives. A glycosylated octadecanoid derivative (palmitoyl dihexoside) could be also identified. In addition to shorter chain fatty acyls such as saturated palmitic acid chain, dodecanedioic acid 38 or ocatanedioic acid and unsaturated nonenoic and dodecadienoic acid 3 alongside with dihexoside decanoic acid derivative were also identified. UPLC-MSn facilitated the tentative identification of fifteen Glycerophospholipids distributed over three main classes (6 glycerophophocholines; PC, 8 glycerophosphoethanolamines; PE and 1 glycerophosphoinositol; PI) with four glycerolipids and one sphingolipid. In addition, flavonoids of different subclasses such are flavone, biflavonoid, polyflavonoid, flavane, chalcone and flavonoid glycosides were putatively identified.
