ZEINAB MOHAMED LABIB;

Abstract

Two hundred and eighteen mature albino rats of both sexes (l 90-200gm b.wt.)

were used in this investigation to study the effect of determination of the oral LD50 (acute toxicity), teratogenicity, and its effect on male fertility as well as pathological examination of the different organs from 1 to 6 months (acute and chronic toxicity).

Experiment No.I determination the oral LD50 (acute toxicity) by Using eighty mature rats of both sexes were used in these study. They were administered Dithane M-45. Toxic symptoms and pathological findings on the tested rats were recorded. After 24 hours of administration, mortality rate and different successive graded doses employed were recorded. The LD50 of dithane was calculated as5187.5 mg/kg/.b. wt

Experiment No 2 studying the teratogenic potency of Di thane M-45 in this experiment 30 mature virgin female rats were housed in ten cages under sanitary condition with 10 males in a ratio of one male to three females. On the day 20th of gestation the female's rats were killed and then the ovary and the uterus were examined and the uterine horns promptly exposed to morphological examination were carried on the pregnant females rats.

Skeletal examination of the eviscerated foeti, which preserved for examination of any abnormalities in the shape or in size and absence of bones. The obtained data clearly demonstrated a great decrease of uterine implantation in treated groups when comparing with control. The percentage of resorped foeti was increased in the treated group compared with control.
