STUDIES ON THE DEVELOPMENT OF A MULTIVALENT VIRAL VACCINE FOR DOGS

Abstract
In a trial aiming to achieve a highly potent, economic and safe trivalent combined vaccine of lyophilized living attenuated Canine distemper
virus and Canine parvo virus vaccine dissolved in an inactivated tissue culture rabies vaccine into one injection and comparing between them. Meanwhile to estimate the level of antibodies status and antigenic value of a given vaccines for evaluation its potentiality. Controlling such diseases, to prevent their spread among dogs and to prevent infection to human. In order to perform the present study, the following steps were performed.
Firstly, prepare batches of each of the three viruses:
-Rabies virus was prepared on BHK-21cell line and titrated before inactivation to give 1075 TCID50 /ml, then inactivated by BEl at a final concentration ofl %, then adjuvated with rehydragel at a final concentration
of20 %.
- Also CPV and CDV was prepared on VERO cells and titrated to
grve 104 5
and 104
TCID50 /ml respectively and then lyophilized after adding
sucrose and lactoalbumin hydrolysate as stabilizer (1:4)
(The sterility test was applied for three batches of vaccines and each batch must be free from any contaminants)
- Then mixing the protective dose ofCPV, CDV and rabies vaccine
which is produced in a suspension and used as a diluent (2m!at injection).
• For evaluation of three mixed vaccines, all vaccines must pass the sterility and safety tests.
