STUDIES ON MICRO PROPAGATION OF SOME OLIVE CULTIVARS

Abstract

This investigation was carried out during (2018-2021) at the tissue culture Lab. of Pomology department, Faculty of Agriculture, Cairo University, Giza, Egypt in cooperation with the National Research Center to establish a micro propagation protocol for mass propagation of three olive cultivars (Picual, Sourani and Dolce). Explant shoots tip was collected from mature, 20-year-old, healthy olive trees for Picual, Sourani and Dolce’ cultivars. During starting stage olive shoots were immersed in silver nanoparticles at different concentrations (100, 200 and 400) mg/L of silver nanoparticles as surface sterilization. Also, olive media (OM) was supplemented with the fresh prepared sliver nanoparticles (AgNPs) at 0, 5, 10 and 20 mg/L. The explants were cultured on different media types, different cytokinin types and concentrations during multiplication stage. Also, different auxin types, concentrations and three different mediums were studied during rooting stage. Darkening of rooting media was performed with 1g L-1 active charcoal. Tissue culture parameters as bud sprouting, shoot growth, multiplication rate and root growth were determined.
