Production and optimizat ion of l -asparaginase by bacteria isolated from Saudi Arabia

Abstract
A total of thirteen isolates were isolated f rom
different areas at Saudi Arabia using M9 agar
medium. Only five out of the 13 isolates were
selected and screened according to their
higher enzyme productivi ty. The strains were
identif ied according to their morphological and
cul tural characteristics, phylogenetic analysis
as well as 16S rRNA Gene Ampl ification and
Sequencing. Optimization of parameters for Lasparaginase
production were achieved af ter
three days incubation periods, 30-40o C
incubation temperature and pH value ranging
f rom 9.0 - 8.0. The enzyme production was
enhanced by mal tose (carbon source),
peptone and ammonium sulphate (ni trogen
sources) and divalent iron (F+2) as a source of
metal ions
