The ameliorating effect of Rutin Hydrate Coadministration with Potassium Bromate on the acini of the parotid salivary glands of Albino rats (Light and transmission electron microscopic study)

Abstract


Aim: investigate the protective effect of Rutin hydrate on parotid gland acini during KBrO3 consumption. Materials and methods: Twenty-four Albino rats were divided into three groups: Group I: received distilled water, Group II: received KBrO3 (20 mg/Kg) twice weekly for four weeks, Group III: was treated as in group II as well as Rutin hydrate (100 mg/kg) once daily for four weeks. Parotid glands were examined by light and Transmission electron microscopes. Area percentage of vacuoles in acini and acinar diameter were measured. Results: Light microscopy of group II showed nuclear pleomorphism and cytoplasmic vacuolations with dilated blood vessels. Some acini showed irregular outlines with hyalinization between them. Group III showed almost normal acinar structure. By transmission electron microscope, group II showed nuclei with variable sizes, shapes and irregular membrane. Cytoplasm showed vacuolations, damaged rough endoplasmic reticulum (RER) cisternae and swollen mitochondria. Acinar cells of group III appeared almost normal with nuclei of comparable size and shape. Cytoplasm showed few cytoplasmic vacuolations, RER and mitochondria appeared normal. By statistical analysis, group II showed a significant increase in area percentage of vacuoles and acinar diameter when compared to group I and group III. Conclusion: Rutin Hydrate coadministration with KBrO3 decreased its damaging effects on the histological picture of parotid glands acini.
