Numerical Sex Chromosome Abberations in Spermatozoa ofSubfertile Men Using FISH

Abstract


Incidence of chromosomal abnonnalities in subfertile males is higher than in fertile males. The majority of cases of infertility are due to oligoasthenospennia. In subfertile males, the rates of sex chromosome aneuploidy are higher than in the nonnal populations thus increasing the risk of producing abnonnal offsprings.

Sex chromosome abnonnalities have also played an important role in several issues. Numerical chromosome abnonnalities represent a major cause in the high incidence of spontaneously aborted embryos. XXY and XYY individuals, who are now able to reproduce through ICSI, produce significantly high levels of XX,YY and XY spennatozoa; increasing the risk of numerical aberrations in tl1eir offsprings.

Sex chromosome aneuploidy is significantly high in oligoasthenospennic patients. The aim of the present study is to detect sex chromosome numerical aberrations in subfertile males using fluorescent in situ hybridization.

FISH was perfonned on the spennatozoa of o!igoasthenospennic patients. Total sex chromosome disomy was reported in I 00% of our Egyptian patients witl1 a mean of 1.775±1.006. XX disomy was found in 20% of patients with results ranging between 0-1% and a mean of 0.2±0.41. YY disomy was found in 15% of patients witl1 arrange of 0-1% and a mean of 0.15±0.366. Finally, XY disomy was found in
100% of patients. The results ranged from 1-4% and showed a mean of 1.425±0.816.


Total sex chromosome disomy and XY disomy showed significant difference between fertile males and oligoasthenospennic patients, while XX and YY disomy were not significantly increased. High incidences of abnonnal spennatozoa and low counts showed a strong correlation with higher incidences of disomy.
