Abstract


The present study aimed to investigate the possible antifibrotic effect of deferiprone (DFP) in modulating liver fibrosis induced immunologically in rat model by concanavalin A (Con A). Additionally, this study aimed to investigate the additional usefulness of DFP to pegylated interferon-alpha (peg IFN-α) and to detect the possible underlying mechanisms. The study was divided into two parts:

Part I: Screening the hepatoprotective dose of DFP against Con A-induced acute hepatotoxicity:

Thirty six male albino rats were divided into six groups (n=6). Group 1 was control rats and injected only with normal saline (0.1 ml/100 gm weight,i.v.), group 2 were given single dose of Con A i.v. dissolved in saline and groups (3, 4, 5 and 6) were administered different doses of DFP dissolved in distilled water (5, 10, 25, 40 mg/kg, oral, respectively), then after 1 h they were injected by Con A. After 24 h from Con A injection, serum samples were collected from each group, then the rats were sacrificed and liver tissues were isolated.
