Abstract


Vitamin B12 is an essential vitamin that is widely used in medical and food industries. Vitamin B12 biosynthesis is confined to few bacteria and archaea, and as such its production relies on microbial fermentation.
The main target of this work is to obtain new genetically modified bacterial strains able to produce higher production of vitamin B12, using some genetic improvement techniques such as mutation by EMS, UV and protoplast fusion.

The results can be summarized as follows:
1. Isolation of 18 bacterial isolates from three different areas (Menofia - Domiatt- Cairo), all isolates were characterized by various microscopic and cultural examinations. Bacterial isolates were tested for growing them on pseudomonas agar base medium, then selection of six isolates had gram stain negative. The isolates P2 and P5 did not given detectable productivity determined so excluded it. The B12 production (4.5, 3, 2.5 and 2 mg/L) were produced by isolates P1, P3, P4 and P11 respectively, they were determined using colorimetric method, and (4, 2.9 and 1.8 mg/L) for isolates P1, P3 and P11 respectively, were determine
