PHYSIOLGICAL STUDIES ON TOMATO PLANT FOR SALT TOLERANCE USING TISSUE CULTURE

Hossam Mohammed Ali Esmaeil;

Abstract

The practical work of this study were carried out in the laboratory of National Agricultural Research Project (NARP), Project No. (109-A) in the Agronomy Dept., Faculty of Agriculture, Zagazig University. Sharkia, Egypt.
:) The experiment have been done on tomato plant
':J:
:!; (Lycopersicon esculentum Beto 86), during the period from 1994

to 1996. The main objectives of the study are the attempt for initiating callus from tomato plants and estimating the physiological behaviour of this calli tissues when grown on a media containing different concentrations of salinity, and the attempts to obtain tomato calli tolerant to salinity. The obtained
,,,. :! results can be summarized as the following :
'

I ;;::

1- The percentage of seeds germinattion have decreased as the salinity concentrations increased.
2- The callus could be induced on MS-media containing a combination between auxin and cytokinin whereas, the auxin (NAA) concentration should be higher than the cytokinin (kinetin) concentration, and it was found that, the best combination between NAA and kinetin for callus initiation from tomato hypocotyl explants were ( 4 tng /L NAA + 0.4 mg/L kinetin).
