GENETIC CONSTRUCTION OF SOME BACTERIAL STRAINS FOR SOME PESTICIDES BIODEGRADATION

GHADA MOSTAFA EL-SAYED MOHAMMED;

Abstract

This study was conducted at Microbial Genetics Department, Genetic Engineering and Biotechnology division, National Research Centre during the period from 2014 to 2018 with the objective of isolation of potent bacterial isolates in pesticides biodegradation, improvement the biodegradation activity by traditional mutation induction, isolation and identification of organophosphorus hydrolase (oph) gene involved in organophosphorus pesticides biodegradation, cloning and expression of oph genes in suitable expression vectors and detection of metabolites resulted from pesticides microbial degradation using GC/MS. Organophosphorus pesticides such as diazinon and chlorpyrifos are moderately toxic to mammals but misuse of these pesticides led to environmental niches contamination. Chemical methods, incineration and landfilling are the most methods used in OPs detoxification but due to their bad effects on the environment; microbial degradation is more safe and cost effective than other chemical and physical methods used in pesticides detoxification
