Neuroprotective effects of Atorvastatin on Doxorubicin-induced cognitive impairment in rats

Abstract


In the current study, the potential neuroprotective effects of ATV against DOX-induced cognitive impairment (chemobrain) were explored through the assessment of ATV-associated antioxidant, anti-inflammatory and anti-apoptotic mediators. Furthermore, the underlying protective mechanisms of ATV against DOX-induced chemobrain were investigated focusing on the potential role of ATV-mediated effects on HO-1 expression and ER response (Figure 17).
The study was divided into two main phases:
1. Screening phase: Screening the neuroprotective dose of ATV against DOX-induced cognitive impairment
A preliminary dose–response screening study was conducted in which rats were randomized into 6 groups and treated for 4 weeks by receiving DOX (2 mg/kg/week, i.p.) and ATV at 3 different doses (5, 10 and 20 mg/kg/day, respectively, P.O., 5 days per week).
The assessed parameters:
i. Behavioral tests
● Passive avoidance test.
